摘要: |
目的 筛选分离纯化延灯滴丸有效成分的最佳树脂,并确定其工艺条件。方法 以有效成分中灯盏乙素的吸附量和解析率为考察指标,采用静态吸附分离法确定适合的大孔吸附树脂;采用动态吸附法确定分离条件。结果 AB-8树脂对延灯滴丸有效成分具有良好的吸附分离性能。其动态分离纯化工艺条件为:延灯滴丸提取物混悬液浓度为8 mg·mL-1,吸附流速为5 mL·min-1,水洗量为13倍柱体积,洗脱剂为60 %乙醇,洗脱量为2倍柱体积,洗脱速度为1 mL·min-1。结论 AB-8树脂分离延灯滴丸有效成分最佳工艺稳定高效,可推广应用于生产。 |

Study on Extraction of Active Constituents of Yandeng Dripping Pills with Macroporous Adsorption Resin |

Abstract: |
OBJECTIVE To establish the separation and enrichment technology for active constituents in Yandeng dripping pills with macroporous adsorption resin. METHODS The adsorption capacity and elution ratio of scutellarin were used as evaluating criteria. The static adsorption and separation were used in investigation of macroporous adsorption resin; the dynamic adsorption and separation were used in studying the condition of adsorption and separation. RESULTS AB-8 had better property of separation and purification and the optimal technological conditions were: the concentration of the sample of Semen deseuraniae was 8 mg·mL-1; the flow velocity of adsorption was 5 mL·min-1; washing water was 13 times of resin volume; the eluting reagent was 60% ethanol(2 times of the resin volume); eluting velocity was 1 mL·min-1. CONCLUSION The technology of separating and purifying active constituents in Yandeng dripping pills with AB-8 is stable and effective, and can be applied to manufacture. |
