目的 测定和比较日本和国产不同工艺制柴苓汤颗粒剂中指标成分柴胡皂苷a的含量。方法 采用HPLC,色谱柱为Kromasil C18(250 mm×4.6 mm,5 μm);流动相为甲醇-水(75∶25);流速0.8 mL·min-1;柱温30 ℃;检测波长为210 nm。结果 柴胡皂苷a线性范围为4.0~15.0 μg,r=0.999 9,加样回收率为100.39%,RSD为0.80%。结论 柴胡皂苷a的含量为日本津村柴苓汤颗粒剂高于国产仿日本工艺制柴苓汤颗粒剂高于国产习用工艺制柴苓汤颗粒剂。本方法简捷、方便、重复性好,准确度高,可用于柴苓汤颗粒剂的质量检测。 |

Determination of the Content of Saikosaponin a in Different Chailing Decoction Granules by HPLC |

Abstract: |
OBJECTIVE To establish a simple method and compare for the determination of the content of Saikosaponin a in different Chailing decoction granules by HPLC. METHODS Kromasil C18 chromatographic column(250 mm×4.6 mm,5 μm)was used.The mobile phase was methanol-water(75∶25).The flow rate was 0.8 mL·min-1, the column temperature was 30 ℃, and the detection wavelength was set at 210 nm.RESULTS The linear relation range of Saikosaporin a was 4.0-15.0 μg( r=0.999 9).The average recovery was 100.39% with the RSD of 0.80%. CONCLUSION Japanese Jincuntang were better than Imitating Japanese method Chailing decoction granules which were better than Chinese ordinary method granule on the content of Saikosaponin a. The method is simple, sensitive, and feasible for the determination of Saikosaponin a and quality control of Chailing decoction granule. |
