重楼皂苷类化合物体外抗生育功效研究

摘要:
目的 筛选并确证重楼皂苷类化合物的抗生育功效。方法 以重楼分离纯化获得的偏诺皂苷(PHAC-A)和薯蓣皂苷(PHAC-B)为实验材料,运用体外孵育小鼠精子测定其活性,研究其对体外精子活性的影响。结果 PHAC-A,PHAC-B体外均具抗生育活性,与阳性对照醋酸棉酚比较,能显著抑制精子活力和精子成活率,且PHAC-B抗生育效果优于PHAC-A,其抗生育活性存在剂量依存关系。结论 PHAC-A,PHAC-B抑制精子活力和精子成活率的功效显著,为高活性抗生育物质,其中PHAC-B为重楼中具抗生育能力的主要活性物质。

Abstract:
OBJECTIVE To study the effect of the saponins of Rhizoma Paridis Yunnanensis [Rhizoma from Paris polyphylla Smith var. yunnanensis(Franch.) Hand.-Mazz.] on male mice procreation. METHODS The pennogenyl saponins(PHAC-A) and dioscins(PHAC-B) was extracted and purified from Rhizoma Paridis Yunnanensis. In the experiment of mice in vitro, the spermicidal effect on male mice of the PHACs in different concentration was measured. RESULTS The spermicidal effect in vitro on male mice of the PHACs was provided with excellent inhibition result in antifertility activities. Compared with gossypol acetic acid, PHACs showed a good inhibition result in spermicidal activities and survival rate, at the same time, PHAC-B had more inhibiting activities than PHAC-A, and had a direct proportion of the activities with their dosage. CONCLUSION Both of the PHACs showed the high inhibitory effect of the sperm activity and survival rate on mice procreation, and they were excellent activitie-compounds in antifertility ability. Compared with PHAC-A and PHAC-B, in Rhizoma Paridis Yunnanensis, the higher activity composition was the PHAC-B.
