豆腐果苷对慢性不可预知温和应激大鼠神经钙调素δ表达的影响

摘要:
目的 研究豆腐果苷对慢性不可预知温和应激(chronic unpredictable mild stress,CUMS)大鼠模型抑郁样行为的作用及对神经钙调素δ(neurocalcin-delta,NCALD)基因在血液中的浓度与前额叶皮质中表达水平的影响,分析NCALD在血液与前额叶皮质中的相关性。方法 连续给予大鼠CUMS与孤养6周,建立大鼠抑郁模型。CUMS大鼠随机分成模型组,盐酸氟西汀组(5 mg·kg-1),豆腐果苷高剂量组(32 mg·kg-1)、中剂量组(16 mg·kg-1)、低剂量组(8 mg·kg-1),继续给予CUMS同时连续灌胃给药6周。通过体质量测量、蔗糖偏嗜度试验、旷场试验评定大鼠行为学变化。Western blotting检测各组大鼠前额叶皮质中NCALD表达水平,ELISA检测血液中NCALD浓度,并探讨两者的相关性。结果 豆腐果苷可使CUMS大鼠体质量减少、蔗糖偏嗜度降低、自主活动减少和探索意愿降低等抑郁行为逆转,可使CUMS大鼠NCALD在血液中浓度升高和前额叶皮质表达上调情况逆转,并且NCALD在血液中浓度与前额叶皮质中表达水平正相关(r=0.771)。结论 豆腐果苷可以改善CUMS导致的抑郁样行为,起到抗抑郁作用,其机制可能与抑制NCALD表达有关。

Effect of Helicid on Neurocalcin-delta Expression in Chronic Unpredictable Mild Stress Rats

Abstract:
OBJECTIVE To study the effect of helicid on the depression-like behavior, and neurocalcin-delta(NCALD) concentration in the blood and expression level in prefrontal cortex in chronic unpredictable mild stress(CUMS) rats, and to analyze the correlation between NCALD concentration in blood and prefrontal cortex. METHODS The CUMS and orphanage were continuously given to rats for six weeks to establish the rat depression model. The CUMS models were then divided into model group, fluoxetine hydrochloride group(5 mg·kg-1) and helicid high, medium, low dose group(32, 16, 8 mg·kg-1). All received CUMS and intragastric administration for another six weeks. The behaviour changes were assessed through body weight, sucrose preference test and open field test. The expression level of NCALD in the prefrontal cortex of each group was detected by Western blotting, and the concentration of NCALD in the blood was detected by ELISA, and the correlation between the two was discussed. RESULTS Helicid could reverse depression behaviors such as weight loss, reduce sucrose bias, voluntary activity, and willingness to explore, and reverse the increase of NCALD concentration in blood and up-regulation in prefrontal cortex expression in CUMS rats. The NCALD concentration in blood was positively correlated with prefrontal cortex expression level(r=0.771). CONCLUSION Helicid has antidepressant effect, and can improve the depression-like behavior caused by CUMS. The mechanism may be related to the inhibition of NCALD expression.
