HPLC测定大风艾中5种二氢黄酮类成分的含量

摘要:
目的 测定大风艾药材中3,3',5,7-四羟基-4'-甲氧基二氢黄酮、3',5,5',7-四羟基二氢黄酮、3,3',4',5-四羟基-7-甲氧基二氢黄酮、3,3',5-三羟基-4',7-二甲氧基二氢黄酮、艾纳香素5种二氢黄酮类成分的含量。方法 采用高效液相色谱法测定,色谱柱为AKZONOBEL Kromasil 100-5C18(4.6 mm×250 mm,5 μm);流动相为乙腈(A)-0.2%冰醋酸(B),梯度洗脱(0~10 min,20% A; 10~15 min,20%→25% A; 15~25 min,25% A; 25~50 min,25%→65% A);流速:1.0 mL·min-1;检测波长:289 nm;柱温:35℃;进样量:10 μL。结果 5种二氢黄酮类成分的线性关系良好,加样回收率为98.73%~101.84%。结论 该方法简便、准确,可用作大风艾药材质量控制的方法。

Simultaneous Determination of Five Flavanones in Blumea Balsamifera by HPLC

Abstract:
OBJECTIVE To determine content of 5 flavanones of 3,3',5,7- tetrahydroxy-4'-methoxyflavanone, 3',5,5',7- tetrahydroxy-flavanone, 3,3',4',5-tetrahydroxy-7-methoxyflavanone, 3,3',5-trihydroxy-4',7-dimethoxyflavanone, 5-blumeatin in Blumea balsamifera. METHODS The chromatographic separation was achieved on AKZONOBEL Kromasil 100-5C18 (4.6 mm×250 mm, 5 μm) column with the mobile phase consisting of acetonitrile(A)-0.2% HAc(B) by gradient elution program(0-10 min, 20%A; 10-15 min, 20%→25%A; 15-25 min, 25%A; 25-50 min, 25%→65%A) at the flow rate of 1.0 mL·min-1, detection wavelength was set at 289 nm, and the column temperature was 35℃, and the injection volume was 10 μL. RESULTS Linearity relationships of 5 flavanones were good, and the average recoveries were from 98.73% to 101.84%. CONCLUSION The method is simple, accurate and repeatable and it is suitable for quality control of Blumea balsamifera.
