基于脂多糖诱导斑马鱼炎症模型研究何首乌多糖抗炎活性

摘要:
目的 探究何首乌中水溶性多糖(water-extractedPolygonum multiflorum polysaccharides,WPMP)对脂多糖(lipopolysaccharide,LPS)所致斑马鱼系统性炎症的保护作用。方法 WPMP经分离纯化后得到2种组分(WPMP-1和WPMP-2),收集斑马鱼胚胎于28.5℃中培养箱中孵育,设置空白组、LPS诱导模型组,通过孵化率、心率、卵黄囊面积、活性氧生成率、NO生成率等指标确定LPS最佳诱导炎症浓度,确定炎症模型后,加入不同组分的WPMP,并以上述相同指标评价WPMP的抗炎效果。结果 不同浓度WPMP (10,50,100 μg·mL-1)对LPS引起的斑马鱼炎症具有保护作用,呈现浓度依赖性,能够恢复卵黄囊正常大小,显著降低心率、活性氧生成率以及NO生成率,且在相同多糖浓度下,WPMP-2的抗炎活性高于WPMP-1。结论 PMP具有较好的抗炎作用,为WPMP作为一种天然的抗炎成分提供了科学依据。

Anti-inflammatory Activity of Polysaccharide from Polygonum Multiflorum Thumb. Based on Lipopolysaccharide Induced Inflammation Model of Zebrafish

Abstract:
OBJECTIVE To investigate the protective effects of water-extracted Polygonum multiflorum polysaccharides (WPMP) on systemic inflammation induced by lipopolysaccharide(LPS) in zebrafish. METHODS Two components of WPMP were isolated and purified(WPMP-1 and WPMP-2). Zebrafish embryos were collected and cultured at 28.5℃. Blank group and LPS-induced model group were set up. The optimal induction concentration of LPS was determined by hatchability, heart rate, yolk sac size, reactive oxygen species(ROS) production rate and NO production rate, and after the establishment of inflammatory model, the anti-inflammatory effects of WPMP were evaluated by the above indexes. RESULTS Different concentration of WPMP(10, 50, 100 μg·mL-1) had a concentration-dependent protective effect on inflammation induced by LPS, and could significantly reduce the size of yolk sac, heart rate, ROS production rate and NO production rate. Under the same polysaccharide concentration, the anti-inflammatory activity of WPMP-2 was higher than that of WPMP-1. CONCLUSION WPMP has good anti-inflammatory effect, which provides scientific basis for WPMP as a natural anti-inflammatory component.
