基于UPLC-Q-TOF-MS和多元统计分析的白术道地性研究

摘要:
目的 对不同产地白术药材的化学成分进行系统分析,为白术的道地性研究提供依据。方法 采用超高效液相色谱-四极杆飞行时间质谱(UPLC-Q-TOF-MS),结合质谱分子网络技术,快速鉴定白术的主要化学成分。应用正交偏最小二乘法-判别分析(OPLS-DA)对安徽、河北、浙江的25批白术进行多元统计分析,用VIP值、t检验筛选出组间差异显著的指标性成分。结果 不同产地的白术化学差异较为明显,共鉴定出10个差异化合物,其中浙江白术的姜黄烯、Atractylmacrol D、白术内酯Ⅲ、3β-羟基苍术酮、白术内酯Ⅱ、白术内酯Ⅰ、β-Vatirenene和苍术酮等成分显著高于安徽或河北样品,可作为潜在的道地性标志物。结论 本研究为全面评价白术药材的质量和道地性提供了依据,并发现不同产地白术的化学成分差异较大,可能与环境、品种和种植方式等有较大关系。

Abstract:
OBJECTIVE To systematically analyze the chemical constituents of Atractylodis Macrocephalae Rhizoma from different origins for providing a basis for its geoherbalism research. METHODS Ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS) combined with mass spectrometry molecular networking was used to rapidly identify the main chemical components of Atractylodis Macrocephalae Rhizoma. Orthogonal partial least squares-discriminant analysis(OPLS-DA) was applied to multivariate statistical analysis of 25 batches of Atractylodis Macrocephalae Rhizoma from Anhui, Hebei and Zhejiang, and the VIP value and t test were used to screen out the index components that differed significantly between groups. RESULTS The chemical differences of Atractylodis Macrocephalae Rhizoma from different origins were obviously. A total of 10 differential compounds had been identified. Among them, the composition of curcumene, Atractylmacrol D, atractylenolide Ⅲ, 3β-hydroxy-atractylon, atractylenolide Ⅱ, atractylenolide Ⅰ, β-Vatirenene and atractylon in Zhejiang Atractylodis Macrocephalae Rhizoma was significantly higher than that in Anhui or Hebei, which could be used as potential geoherbalism markers. CONCLUSION This study provides a basis for a comprehensive evaluation of the quality and geoherbalism of Atractylodis Macrocephalae Rhizoma and finds that the chemical composition of Atractylodis Macrocephalae Rhizoma varies considerably among different origins, which may be related to the environment, variety and cultivation methods, etc.
