摘要: |
目的:研究杜钩平肝颗粒处方的制备工艺、质量标准及初步药效学,为研制成颗粒剂提供基础。方法:采用正交实验法进行水提工艺研究,以出膏率及葛根素的含量为指标,优选出最佳的水提工艺。采用薄层鉴别及高效液相色谱法对杜钩平肝颗粒进行质量标准研究。研究杜钩平肝颗粒对L-NAME诱导高血压大鼠模型的预防作用。结果:杜钩平肝颗粒处方最佳的提取工艺为:10倍量的水,浸泡1h,煎煮1.5h,煎煮3次。质量标准为:以葛根素作为含量测定指标,以盐酸小檗碱、川牛膝、盐酸水苏碱作为鉴别指标。药效学试验结果表明,杜钩平肝颗粒低剂量组能降低大鼠收缩压和平均动脉压,杜钩平肝颗粒高剂量组能降低大鼠收缩压。结论:杜钩平肝颗粒制备工艺稳定可行,且具有一定的降压作用,为医疗机构制剂的备案提供了药学基础。 |

Objective: To study the preparation process, quality standards, and preliminary pharmacodynamics of Dugoupinggan Granules, providing a basis for the preparation of medical institutions. Methods: The orthogonal experimental method was used to study the water extraction process, and the optimal water extraction process was selected based on the extraction rate and the content of puerarin. The quality standard of Duguopingan Granules was studied by TLC and HPLC. To study the preventive effect of Dugoupinggan Granule on L-NAME induced hypertension in rats. Results: The optimal extraction process for Dugoupinggan Granules is: 10 times the amount of water, soaking for 1 hour, decocting for 1.5 hours, and decocting for 3 times. The quality standard uses puerarin as the content determination index, and berberine hydrochloride, achyranthes bidentata, and stachydrine hydrochloride as the identification index. The pharmacodynamic test results showed that the low dose group of Dugoupinggan Granule could reduce systolic blood pressure and mean arterial pressure in rats, while the high dose group of Dugoupinggan Granule could reduce systolic blood pressure in rats. Conclusion: The preparation process of Dugoupinggan Granules is stable and feasible, and has a certain antihypertensive effect, providing a pharmaceutical basis for the filing of preparations in medical institutions. |
