目的 探讨10批不同来源米槁挥发油对心肌损伤大鼠的治疗作用,并结合熵权法对比分析不同来源米槁挥发油对心肌损伤大鼠的治疗活性。方法 建立心肌损伤大鼠模型,采用蛋白免疫印迹法(Western blotting,WB)检测心肌组织中Bax/Bcl-2蛋白,实时荧光定量PCR技术(Real-time Quantitative PCR,RT-qPCR)检测心肌组织中成纤维细胞生长因子2(FGF-2)水平,免疫组织化学法(mmunochemistry,IHC)分析10批不同来源的米槁挥发油治疗心肌损伤大鼠的心肌及主动脉组织的纤溶酶原激活物抑制剂-1(plasminogen activator inhibitor 1,PAI-1)及组织型纤溶酶原激活物(tissue plasminogen activator,TPA)的表达,苏木素-伊红染色法(Heatoxylin-eosin staining,HE)检测心肌组织的病变情况,通过熵权法综合评价10批不同来源米槁挥发油对心肌损伤大鼠的治疗效果。结果 10批不同来源的米槁挥发油均可使心肌组织中Bax/Bcl-2水平降低,FGF-2水平降低,心肌和主动脉组织的PAI-1水平降低并升高TPA水平,缓解心肌组织的病变,熵权法计算结果以心肌组织中TPA的权重最大,贵州罗甸双江镇(2019年10月)、册亨县林木村(2019年10月)米槁挥发油综合评分最高,贵州罗甸罗悃镇(2018年9月)、罗甸罗悃镇(2018年10月)米槁挥发油评分最低。结论 10批不同来源的米槁挥发油可能通过调节Bax/Bcl-2蛋白的表达,降低FGF-2水平抑制心肌肥厚和心脏纤维化,增加纤溶系统阻止纤溶蛋白在血管沉积和血栓形成并保护心肌组织,从而实现对心肌损伤大鼠的治疗作用,其中不同来源米槁挥发油的治疗能力存在一定差异,以贵州罗甸双江镇(2019年10月)、册亨林木村(2019年10月)米槁挥发油治疗效果相对较优,贵州罗甸罗悃镇(2018年9月)、罗甸罗悃镇(2018年10月)米槁挥发油治疗效果相对较差,可为后续米槁挥发油治疗心肌损伤的研发提供参考。 |

OBJECTIVE To investigate the therapeutic effect of 10 batches of volatile oil from different sources of Cinnamomum migao H.W.Li on rats with myocardial injury and to compare and analyze the therapeutic activity of volatile oil from different sources of Cinnamomum migao H.W.Li on rats with myocardial injury in combination with entropy weight method. METHODS A rat model of myocardial injury was established, and Bax/Bcl-2 protein in myocardial tissue was detected by Western blotting (WB), and the level of fibroblast growth factor-2(FGF-2) in myocardial tissue was detected by quantitative real-time PCR (RT-QPCR). Immunohistochemistry (IHC) was used to analyze the plasminogen activator inhibitor-1 (Plasminogen activator inhibitor 1, The expressions of PAI-1 and tissue plasminogen activator,TPA) were detected by hematoxylin-eosin staining (He), and the weight obtained by entropy weight method was used as the resolution coefficient to evaluate the therapeutic effect of 10 batches of Cinnamomum migao H.W.Li volatile oil from different sources on myocardial injury rats. RESULTS Ten batches of Cinnamomum migao H.W.Li volatile oil from different sources can reduce the levels of Bax/Bcl-2, FGF-2, PAI-1 and TPA in myocardium and aorta, thus alleviating the pathological changes of myocardium. The results calculated by entropy weight method show that TPA is the most important in myocardium, with shuang jiang in Luodian, Guizhou (October 2019) and Linmu Village in Ceheng County (October 2019). CONCLUSION Ten batches of Cinnamomum migao H.W.Li volatile oil from different sources may have therapeutic effects on myocardial injury rats by regulating the expression of Bax/Bcl-2 protein, reducing FGF-2 level, increasing fibrinolytic system, preventing fibrinolytic protein from depositing in blood vessels and thrombosis, and protecting myocardial tissue. Among them, the therapeutic ability of Cinnamomum migao H.W.Li volatile oil from different sources is different. The therapeutic efficacy of Cinnamomum migao H.W.Li volatile oil in Shuangjiang Town, Luodian, Guizhou (October 2019) and Linmu Village, Ceheng (October 2019) was relatively better, and the therapeutic efficacy of Cinnamomum migao H.W.Li volatile oil in Luodian Luoxiao Town, Luodian, Guizhou (September 2018) and Luoxiao Luoxiao Town, Luodian, Guizhou (October 2018) was relatively poorer, which may provide a reference for the research and development of the subsequent Cinnamomum migao H.W.Li volatile oil in the treatment of myocardial injury. |
