| 目的 建立黄草乌药材的含量测定方法,提升黄草乌药材的质量控制方法。方法 通过显微鉴别、薄层鉴别进行定性鉴别;通过水分、总灰分、酸不溶灰分、浸出物检测药材质量;采用HPLC法检测滇乌碱与黄草乌碱甲的含量。结果 黄草乌显微特征明显;薄层色谱斑点清晰,分离度较好;水分6.35~10.65%,总灰分2.16~4.81%,酸不溶灰分0.0649~0.8032%,浸出物37.10~52.38%;滇乌碱与黄草乌碱甲在40~2000 ng范围内线性关系良好,滇乌碱含量0.0106~0.0554%,黄草乌碱甲含量0.0051~0.0502%,滇乌碱与黄草乌碱甲总含量0.0239~0.0950%。结论 该方法合理可靠,可用于黄草乌药材的质量控制。 |

| OBJECTIVE To establish an HPLC method for the content determination and to enhance the quality control method for Radix Aconitum Vilmoriniani. METHODS Qualitative identification was carried out by microscopic identification and thin-layer identification, and the quality of the medicinal materials was assessed by moisture, total ash, acid-insoluble ash, and leachate. The content of yunaconitine and vilmorrianine A in raw materials were determined by HPLC method. RESULTS The microscopic characteristics of Radix Aconitum Vilmoriniani are distinct, with clear spots and good resolution in thin-layer chromatography. The moisture content of the raw materials ranges from 6.35% to 10.65%, total ash from 2.16% to 4.81%, acid-insoluble ash from 0.0649% to 0.8032%, and leachate from 37.10% to 52.38%. The linearity correlations for yunaconitine and vilmorrianine A are well within the range of 40~2000 ng. The content of yunaconitine varies between 0.0106% and 0.0554%, while the content of vilmorrianine A ranges from 0.0051% to 0.0502%. The combined content of yunaconitine and vilmorrianine A is between 0.0239% and 0.0950%. CONCLUSION This method is reasonable and reliable for the quality control of Radix Aconitum Vilmoriniani. |
