基于多指标-响应面法优选土炒白术的炮制工艺[1]

摘要:
目的 对土炒白术的炮制工艺进行优化。方法 采用紫外可见分光光度法和超高效液相色谱-三重四极杆串联质谱(UPLC-QqQ-MS/MS)技术对土炒白术中多糖含量以及白术内酯Ⅰ、白术内酯Ⅱ、白术内酯Ⅲ等活性成分的含量进行测定,采用色差仪对土炒白术粉末色度值进行分析;将上述化学成分的含量以及饮片的色度作为评价指标,采用单因素试验法对土炒白术过程中的辅料用量、炒制温度、炒制时间进行考察;通过层次分析-赋值(analytic hierarchy process and criteria importance through inter criteria correlation,AHP-CRITIC)混合加权法结合Box-Behnken响应面法进行土炒白术炮制工艺的优化研究。结果 优化土炒白术的最佳炮制工艺为每50 g药材用灶心土28.4 g,在204 ℃下炒制5.30 min。结论 本实验确定的土炒白术工艺切实可行,可为该炮制品的工业化生产中提供一定的指导意义。

Abstract:
Objective Optimize the processing technology of soil stir-fried AMR. Methods By using Uv-Visible Spectrophotometry and Ultra-Performance Liquid Chromatography-Triple Quadrupole Tandem Mass Spectrometry (UPLC-QqQ-MS/MS) to determine the content of polysaccharides and active ingredients such as atractylenolideⅠ, atractylenolideⅡ, atractylenolide Ⅲ etc. The colorimeter was used to determine the color value of soil-fried AMR powder. The contents of the above chemical components and the color value of soil-fried AMR pieces were used as evaluation indexes. The single factor trial method was adopted to investigate the amount of excipients,,stir-frying temperature and time of AMR. The analytic hierarchy process and criteria importance through inter criteria correlation (AHP-CRITIC) combined with the method of Box-Behnken response surface to design and optimize the processing technology of soil stir-fried AMR. Results The best processing technology of soil fried AMR were optimized to 28.4 g of soil added to 50 g of medicinal materials which was fried 5.30 min in 204℃. Conclusion The process of soil stir-fried AMR determined in this experiment is feasible which can provide certain guiding significance for the industrial production of the processed products.
