| 目的? 从油茶内生真菌Diaporthe arengae M2的次级代谢物中寻找抑菌活性先导化合物。方法? 采用传统柱层析法以及semi-prep HPLC对D. arengae M2次级代谢物进行分离纯化,再通过NMR、IR、HR-ESI-MS等波谱仪器鉴定化合物结构,最后使用肉汤稀释法对这些化合物进行抑菌活性评价。结果? 从D. arengae M2的次级代谢物中分离鉴定了8个化合物:xylariphthalide A (1)、cytosporone B (2)、cytosporone E (3)、cytosporone C (4)、pestalotiopsone A (5)、pestalotiopsone C (6)、ergosterol (7)、7, 8-dimethyl-3, 10-dimethyl-iso-alloxazine (8)。抑菌活性评价显示,化合物3对3株革兰氏阳性菌的MIC值均为2 μg/mL,对E.coli与P. aeruginosa的MIC值分别为32与64 μg/mL;化合物2对4株尖孢镰刀菌的MIC值的范围在8 – 16 μg/mL之间;化合物4可以抑制S. aureus、MRSA与B.subtilis,MIC范围在8 - 16 μg/mL之间。结论? 化合物2,3和4具有良好的抑菌活性。 |

| ABSTRACT: OBJECTIVE To find lead compounds of antibacterial activity from secondary metabolites of endophytic fungus Diaporthe arengae M2 in Camellia oleifolia. METHODS The secondary metabolites of D. arengae M2 were isolated and purified by traditional column chromatography and semi-prep HPLC. The structure of the compounds was identified by NMR, IR, HR-ESI-MS and other spectroscopic instruments. Finally, the antibacterial activity of these compounds was evaluated by broth dilution method. RESULTS A total of 8 compounds were isolated and identified from the secondary metabolites of D. arengae M2: xylariphthalide A (1), cytosporone B (2), cytosporone E (3), cytosporone C (4), pestalotiopsone A (5), pestalotiopsone C (6), ergosterol (7), 7, 8-dimethyl-3, 10-dimethyl-iso-alloxazine (8). The antibacterial activity evaluation showed that the MIC values of compound 3 against the three Gram-positive strains were 2 μg/mL, and the MIC values against E.coli and P. aeruginosa were 32 and 64 μg/mL respectively. The MIC values of compound 2 against four strains of Fusarium oxysporum ranged from 8 to 16 μg/mL. compound 4 against S. aureus, MRSA and B.subtilis with MIC ranged from 8 to 16 μg/mL. CONCLUSION Compounds 2, 3 and 4 showed better antibacterial activity. |
