基于UPLC-Q-TOF-MS/MS技术比较桂枝-甘草药对单煎液和合煎液的入血成分

摘要:
对桂枝-甘草药对单煎液和合煎液的入血成分进行分析比较,为桂枝-甘草药对的药效物质基础研究提供参考。方法 通过UPLC-Q-TOF-MS/MS技术,根据提取离子流色谱图、质谱碎片裂解信息识别目标化合物,结合TCMSP、BATMAN-TCM以及知网等综合性数据库,筛选入血成分。对桂枝、甘草以及药对含药血清的化学成分进行分析。结果 桂枝、甘草及其药对均含有丰富的化学成分。从桂枝、甘草及其药对含药血清中分别鉴定出103个、164个、143个化学成分。其中桂枝入血原型成分20个,代谢产物83个;甘草含药血清入血原型成分41个,潜在入血原型成分代谢产物123个;桂枝-甘草药对含药血清入血原型成分31个,代谢产物112个;主要入血成分包括黄酮类、酚类、苯丙素类及萜类。结论 本研究对桂枝-甘草单煎液和合煎液入血成分进行了定性分析,明确了其入血成分的差异。为桂枝-甘草药对的药效物质基础和作用机制的进一步研究提供参考。

Abstract:
The blood components of Gui Zhi-Gan Cao medicinal pairs were analysed and compared with those of single decoction and combined decoction, so as to provide a reference for the basic research on the pharmacological substances of Gui Zhi-Gan Cao medicinal pairs. METHODS The target compounds were identified by UPLC-Q-TOF-MS/MS technique based on extracted ion flow chromatograms, mass spectrometry fragmentation cleavage information, and screened for haematological constituents by combining comprehensive databases such as TCMSP, BATMAN-TCM, and CNKI Networks. The chemical composition of Guizhi-Gancao herb pair containing serum was analysed. RESULTS The blood components of Gui Zhi,Gan Cao, and their medicinal pairs are rich in chemical constituents. Total 103, 164, 143 constituents were identified from the blood serum of Gui Zhi, Gan Cao, and Gui Zhi-Gan Cao medicinal pairs, respectively. Gui Zhi plasma samples containing 20 potential prototype blood components, and 83 metabolites of potential prototype blood components. Gan Cao plasma samples containing 41 potential prototype blood components, and 123 metabolites of potential prototype blood components.Gui Zhi-Gan Cao medicinal pairs plasma samples containing 31 potential prototype blood components, and 112 metabolites of potential prototype blood components. Mainly differentiated including flavonoids, phenolics, phenylpropanoids, alkaloids, and other components. CONCLUSION In this study, the blood-entry components of Gui zhi-Gan cao single decoction and combined decoction were qualitatively analysed, and the differences in blood-entry components were clarified. It provides a reference for further studies on the pharmacological material basis and mechanism of action of Gui Zhi-Gan Cao medicinal pairs.
