基于物理、化学指纹图谱及多成分含量测定构建养肝降草汤质量评价方法

摘要:
目的 建立15批养肝降草汤的物理、化学指纹图谱,并对其7种指标成分进行含量测定,采用“物理指纹图谱+化学指纹图谱+含量测定”评价体系全面表征该汤剂的整体质量。方法 制备15批养肝降草汤,以动力黏度、折光率、电导率、溶解性固体总量、固含量、密度及pH值7个物理参数构建物理指纹图谱并利用相似度评价、层次聚类分析及主成分分析对物理指纹图谱进行化学模式识别分析;以HPLC法对红景天苷、栀子苷、连翘酯苷A、新橙皮苷、黄芩苷、甘草酸、白术内酯III 7种指标成分进行含量测定并构建化学指纹图谱,结合物理指纹图谱综合评价汤剂的整体质量。结果 15批汤剂物理指纹图谱之间相似度大于0.89, HCA结果显示,15批汤剂聚为2类;化学指纹图谱中15批汤剂与对照图谱相比相似度均大于0.90; 15批汤剂中7种指标成分含量为红景天苷0.208 4~ 0.253 7 mg?g-1、栀子苷2.030 4~ 2.426 3 mg?g-1、连翘酯苷A 0.774 6~ 0.950 1 mg?g-1、新橙皮苷0.758 1~ 1.239 2 mg?g-1、黄芩苷4.777 2~ 5.941 5 mg?g-1、甘草酸0.147 8~ 0.234 8 mg?g-1、白术内酯III 0.007 9~ 0.016 6 mg?g-1含量HCA结果除BZTJ5外,其余批次与物理指纹图谱HCA结果一致。结论 结果表明养肝降草汤的物理参数与化学成分具有相关性,可以将物理指纹图谱与化学指纹图谱相结合,用于更精准把握养肝降草方制剂的整体质量。

Abstract:
Objective To establish the physical fingerprints and HPLC fingerprints of 15 batches of decoction, and the content of 7 index components was determined, and to the evaluate overall quality of the decoction by multiple means. Methods Fifteen batches of decoction were prepared, and the physical fingerprint was constructed with seven physical parameters, including dynamic viscosity, refractive index, conductivity, total dissolved solids, solid content, density and pH value, and the chemical pattern recognition analysis of the physical fingerprint was carried out by similarity evaluation, hierarchical cluster analysis and principal component analysis; The contents of salidroside, geniposide, forsythoside A, neohesperidin, baicalin, glycyrrhizic acid and atractylenolide III were determined by HPLC and the chemical fingerprint was constructed. Combined with the physical fingerprint, the overall quality of the decoction was comprehensively evaluated. RESULTS The similarity between the physical fingerprints of 15 batches of decoction was greater than 0.89, and the results of HCA showed that the 15 batches of decoction were clustered into two categories. In the chemical fingerprint, the similarity of the 15 batches of decoction was greater than 0.90 compared with the control spectrum. The content of 7 index ingredients in 15 batches of decoction is: salidroside 0.208 4~ 0.253 7 mg?g-1, geniposide 2.030 4~ 2.426 3 mg?g-1, forsythoside A 0.774 6~ 0.950 1 mg?g-1, neohesperidin 0.758 1~ 1.239 2 mg?g-1, baicalin 4.777 2~ 5.941 5 mg?g-1, glycyrrhizic acid 0.147 8~ 0.234 8 mg?g-1and atractylenolide III 0.007 9~ 0.016 6 mg?g-1. Except for BZTJ5, the results of HCA were consistent with the results of physical fingerprint HCA. CONCLUSION The results showed that the physical parameters of the decoction of Yanggan Herb Recipe were correlated with the chemical composition, and the physical fingerprint and chemical fingerprint could be combined to more accurately grasp the overall quality of the Liver Herb Formula.
