| 摘要:
目的:巨噬细胞泡沫化是动脉粥样硬化(AS)发生发展主要原因。探讨牛膝水提物对ox-LDL诱导巨噬细胞泡沫化的作用和机制。方法:在考察牛膝水提物给药浓度的基础上,Raw264.7巨噬细胞分为空白组、ox-LDL模型组、牛膝水提物低中高三剂量组,模型组和牛膝水提物各给药组细胞采用100 μg/mL的ox-LDL诱导形成泡沫化细胞。采用油红O染色和DiI-ox-LDL干预考察细胞脂质沉积和摄取ox-LDL能力;采用Western blot、RT-PCR检测细胞LXRα、ABCG1、SR-BI等蛋白和mRNA表达水平。并采用UPLC-QTOF-MS/MS检测牛膝水提物中成分。结果:牛膝水提物含有阿魏酸、竹节参皂苷Ⅳa等成分。与空白组相比,模型对照组形成了巨噬细胞源性泡沫细胞,表现为脂质沉积和摄取ox-LDL能力明显增加,细胞LXRα、ABCG1蛋白和mRNA表达水平降低。与模型对照组相比,牛膝水提物各剂量可明显降低泡沫化细胞脂质沉积和摄取ox-LDL能力(p<0.01, 0.05);牛膝水提物还可明显增加细胞LXRα、ABCG1蛋白和mRNA表达水平(p<0.01, 0.05)。结论 牛膝水提物可能通过上调LXRα/ABCG1抑制ox-LDL诱导巨噬细胞泡沫化,从而防治动脉粥样硬化。 |

Objective: Macrophage foam is the main cause of atherosclerosis (AS). To explore the effect and mechanism of Achyranthes bidentata Bl. water extract on ox-LDL induced macrophage foam. Method: On the basis of investigating the concentration of Achyranthes bidentata Bl. water extract, Raw264.7 macrophages were divided into blank group, ox-LDL model group, and Achyranthes bidentata Bl. water extract low, medium, and high dose group. The model group and Achyranthes bidentata Bl. water extract each treatment group were treated with 100 μg/mL ox-LDL induced the formation of foam cells. Firstly, oil red O staining and DiI-ox-LDL intervention were used to investigate the ability of cell lipid deposition and uptake of ox-LDL. And Western blot and RT-PCR were used to detect cell LXRα, ABCG1, and SR-BI protein and mRNA expression levels.And UPLC-QTOF-MS/MS was used to detect the components in the Achyranthes bidentata Bl. Result: The Achyranthes bidentata Bl contains components ferulic acid and Chikusetsusaponin IVa.Compared with the blank group, the model control group formed macrophage derived foam cells, which showed that the ability of lipid deposition and uptake of ox-LDL increased significantly, and the cell LXRα, and the expression levels of ABCG1 protein and mRNA decreased. Compared with the model control group, each dose of Achyranthes bidentata Bl. water extract can significantly reduce the ability of foam cells to deposit lipid and absorb ox-LDL (p<0.01, 0.05); Achyranthes bidentata Bl. water extract can significantly increase cell LXRα, and the expression levels of ABCG1 protein and mRNA (p<0.01, 0.05). Conclusion: The Achyranthes bidentata Bl. water extract may upregulate LXRα/ABCG1 inhibits ox-LDL induced macrophage foam, thereby preventing atherosclerosis. |
