| 探究除湿胃苓汤调节AKT/mTOR信号通路对银屑病模型小鼠皮损的改善作用。方法:BALB/c小鼠随机分为正常组、银屑病模型组、除湿胃苓汤低剂量组、除湿胃苓汤中剂量组、除湿胃苓汤高剂量组、除湿胃苓汤高剂量+SC79组,通过涂抹5%咪喹莫特乳膏来构建银屑病模型,采用除湿胃苓汤和AKT激活剂SC79分组干预后进行挠痒行为学、皮肤增厚与皮损症状检测;通过HE染色进行皮肤组织病理形态检测,比较各组小鼠皮肤表皮厚度;以Masson染色检测各组小鼠皮肤组织胶原纤维占比;以ELISA测量各组小鼠血清炎性因子水平;以免疫印迹法检测各组小鼠皮肤组织AKT/mTOR信号通路蛋白表达。结果:相比正常组,银屑病模型组小鼠挠痒次数、皮肤厚度、PASI、皮肤组织皮肤表皮厚度与胶原纤维占比、血清IL-6、IL-17与IL-22水平、皮肤组织p-AKT/AKT与p-mTOR/mTOR升高(P<0.05);相比银屑病模型组,除湿胃苓汤低、中、高剂量组小鼠挠痒次数、皮肤厚度、PASI、皮肤组织皮肤表皮厚度与胶原纤维占比、血清IL-6、IL-17与IL-22水平、皮肤组织p-AKT/AKT与p-mTOR/mTOR降低(P<0.05),并呈剂量依赖性(P<0.05);相比除湿胃苓汤高剂量组,除湿胃苓汤高剂量+SC79组小鼠挠痒次数、皮肤厚度、PASI、皮肤组织皮肤表皮厚度与胶原纤维占比、血清IL-6、IL-17与IL-22水平、皮肤组织p-AKT/AKT与p-mTOR/mTOR升高(P<0.05)。结论:除湿胃苓汤可通过抑制AKT/mTOR信号激活而减轻银屑病模型小鼠炎症与其皮损症状。 |

| To investigate the improvement effect of Chushi Weiling Tang on psoriasis model mice skin lesions by modulating AKT/mTOR signaling pathway. Methods: BALB/c mice were stochastically grouped into normal group, psoriasis model group, low-dose Chushi Weiling Tang group, medium-dose Chushi Weiling Tang group, high-dose Chushi Weiling Tang group, and high-dose Chushi Weiling Tang+SC79 group. The psoriasis model was constructed by applying 5% imiquimod cream. After intervention with Chushi Weiling Tang and AKT activator SC79, the scratching behavior, skin thickening, and skin lesion symptoms were detected. HE staining was used to test the pathological morphology of skin tissue, and the skin epidermal thickness was compared. Masson staining was used to measure the proportion of collagen fibers in the skin tissue. ELISA was used to measure serum inflammatory factors. Immunoblotting was used to detect AKT/mTOR signaling pathway proteins in the skin tissues of mice. Results: For the normal group, the psoriasis model group had increased scratching frequency, skin thickness, PASI, skin epidermal thickness, proportion of collagen fibers in skin tissue, serum IL-6, IL-17 and IL-22, and skin tissue p-AKT/AKT and p-mTOR/mTOR (P<0.05). For the psoriasis model group, the low, medium, and high dose Chushi Weiling Tang groups had decreased scratching frequency, skin thickness, PASI, skin epidermal thickness, proportion of collagen fibers in skin tissue, serum IL-6, IL-17 and IL-22, and skin tissue p-AKT/AKT and p-mTOR/mTOR, in a dose-dependent manner (P<0.05). For the high-dose Chushi Weiling Tang group, the high-dose Chushi Weiling Tang+SC79 group had raised scratching frequency, skin thickness, PASI, skin epidermal thickness, proportion of collagen fibers in skin tissue, serum IL-6, IL-17 and IL-22, and skin tissue p-AKT/AKT and p-mTOR/mTOR (P<0.05). Conclusion: Chushi Weiling Tang can alleviate inflammation and skin lesion symptoms in psoriasis model mice by suppressing AKT/mTOR signaling activation. |
