中药调控Keap1/Nrf2/ARE信号通路干预肺纤维化的研究进展

摘要:
肺纤维化(pulmonary fibrosis,PF)是一种慢性进行性的呼吸系统疾病,其发病率会随着年龄的增长不断增加。氧化应激损伤是肺纤维化的重要发病机制,在肺纤维化的发生发展中具有重要作用。Kelch样ECH相关蛋白1(Keap1)/核转录因子E2相关因子2(Nrf2)/抗氧化反应元件(ARE)信号通路是细胞内重要的抗氧化应激途径之一,该通路可以通过调节多种抗氧化酶和解毒酶的表达,对细胞的氧化还原平衡及代谢起到关键作用。研究发现,中药活性成分、提取物及其复方可通过抑制氧化应激损伤,减轻炎症反应等途径发挥抗肺纤维化的作用,其作用机制与激活Keap1/Nrf2/ARE信号通路有关。基于此,该文从中药调控Keap1/Nrf2/ARE信号通路治疗肺纤维化的作用机制进行综述,为中药治疗肺纤维化的新药研究提供参考。

Abstract:
Pulmonary fibrosis (PF) is a chronic progressive respiratory disease, the incidence of which increases with age. Oxidative stress damage is an important pathogenesis of pulmonary fibrosis and plays a significant role in its development. The Kelch-like ECH-associated protein 1 (Keap1)/nuclear factor E2-related factor 2 (Nrf2)/antioxidant response element (ARE) signaling pathway is one of the key intracellular antioxidant stress pathways. It regulates the expression of various antioxidant and detoxifying enzymes, playing a crucial role in maintaining cellular redox balance and metabolism. Studies have shown that active components, extracts, and formulations of traditional Chinese medicine (TCM) can exert anti-pulmonary fibrosis effects by inhibiting oxidative stress damage and reducing inflammatory responses, with mechanisms related to the activation of the Keap1/Nrf2/ARE signaling pathway. Based on this, this article reviews the mechanisms of TCM regulation of the Keap1/Nrf2/ARE signaling pathway in the treatment of pulmonary fibrosis, providing references for the development of new TCM drugs for the treatment of pulmonary fibrosis.
