基于近红外光谱技术的摩罗丹水提液浓缩过程的多指标快速检测

摘要:
目的 基于近红外光谱(near infrared spectroscopy,NIR)技术,以芍药内酯苷、芍药苷、密度和固含物为质量控制指标,对摩罗丹水提液的浓缩过程进行快速检测,实现摩罗丹浓缩过程在线质量控制。方法 采集摩罗丹水提液浓缩过程的在线近红外光谱,并收集样本;基于HPLC、烘干法等理化仪器分析方法,测定摩罗丹水提液重要指标的含量;然后运用偏最小二乘法建立重要指标的近红外光谱校正模型;最后验证校正模型的性能,并运用校正模型预测摩罗丹水提液浓缩过程重要指标含量的变化。结果 所建立的芍药内酯苷、芍药苷、密度和固含物校正模型的相关系数(R)均 > 0.98,验证集的R均 > 0.99,且验证集相对偏差均 < 7%。结论 近红外光谱分析技术可实现摩罗丹水提液浓缩过程中芍药内酯苷、芍药苷、密度和固含物4个关键质控指标的快速检测,方法能够满足中药实际生产过程分析的准确性要求。

Multi-indicators Rapid Determination in Concentration Process of Moluodan Water Extration Liquid by Near-infrared Spectroscopy

Abstract:
OBJECTIVE Based on near infrared spectroscopy(NIR) spectroscopy technology, taking alibiflorin, paeoniflorin, density and soluble solid content as quality control indicators, to carry out rapid detection of Moluodan aqueous extract in the concentration process, so as to realize the on-line quality control of Moluodan concentration process.METHODS Collect the on-line NIR spectrum of Moluodan aqueous extract in the concentration process, and collect samples. Based on HPLC, drying method and other physical and chemical instrument analysis methods, the content of important indexes of Moluodan aqueous extract were determined. Then the NIR spectrum correction model of important indexes was established by using partial least square method. Finally, the performance of the correction model was verified, and the correction model was used to predict the changes of important indexes in the concentration process of Moluodan aqueous extract.RESULTS The correlation coefficients(R) of the established calibration models for alibiflorin, paeoniflorin, density and solid content were > 0.98, the R of the validation set was > 0.99, and the relative deviation of the validation set was < 7%.CONCLUSION NIR spectral analysis technology can realize the rapid detection of alibiflorin, paeoniflorin, density and solid content in the concentration process of Moluodan aqueous extract, and the results are accurate and reliable.
