UPLC同时测定不同产地三叶青不同部位中的7种活性成分

摘要:
目的 建立UPLC同时测定不同产地三叶青不同部位中7种活性成分含量。方法 采用UPLC分离检测,色谱柱为BEH Shield RP18(150 mm×2.1 mm,1.7 μm);柱温35 ℃;进样量2 μL;流动相:乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱;流速0.3 mL·min-1;检测波长为360 nm和280 nm,采用外标法测定7种活性成分的含量。结果 儿茶素、原花青素B1、芦丁、异槲皮素、山奈酚-3-O-芸香糖苷、紫云英苷、芹菜素分别在1.02~40.8,1.43~57.0,1.16~46.2,1.09~43.6,1.12~44.6,1.18~47.2,0.30~11.8 μg·mL–1呈良好的线性关系。不同产地三叶青和不同部位三叶青中各成分含量均有一定差异。结论 采用UPLC同时测定三叶青中7种活性成分准确可行,为三叶青药材的质量控制提供了参考。

Simultaneous determination of 7 active ingredients from Different Parts of Tetrastigma hemsleyanum of different origins by UPLC

Abstract:
OBJECTIVE To establish a UPLC method for the simultaneous determination of 7 active ingredients in different parts of Tetrastigma hemsleyanum of different origins. METHODS UPLC was used for separation and detection. The chromatographic analysis was performed on BEH Shield RP18 column(150 mm×2.1 mm, 1.7 μm). The column temperature was 35℃. The injection volume was 2 μL. The mobile phase was acetonitrile(A)-0.1% phosphoric acid solution(B) at the flow rate of 0.3 mL·min-1 with gradient elution. The detection wavelengths were 360 nm and 280 nm. The contents of 7 kinds of active ingredients were determined by external standard method. RESULTS Catechin, procyanidin B1, rutin, isoquercetin, kaempferol-3-o-rutoside, astragaloside and apigenin showed a good linear relationship in 1.02-40.8, 1.43-57.0, 1.16-46.2, 1.09-43.6, 1.12-44.6, 1.18-47.2, 0.30-11.8 μg·mL-1, respectively. There were some differences in the contents of various componennts in different areas and different parts of T. Hesleyanur. CONCLUSION The established UPLC method for the simultaneous determination of 7 active ingredients in Tetrastigma hemsleyanum is accurate and feasible, which provides a reference for the quality control of tetrastigmae radix.
