HPLC测定野马追中野马追内酯A,B和金丝桃苷的含量

摘要:
目的 采用高效液相色谱法对不同产地的6批野马追药材中野马追内酯A,B和金丝桃苷进行了定性定量分析。方法 野马追内酯A,B的测定采用Agilent Extend-C18(4.6 mm×150 mm,5 μm)为分析柱,乙腈-水(27∶73)为流动相,流速为1.0 mL·min-1,等度洗脱,检测波长为220 nm,柱温为30 ℃。金丝桃苷的测定以SB-C18为分析柱,乙腈-1%醋酸水溶液(12∶88)为流动相,流速为1.0 mL·min-1,等度洗脱,检测波长为255 nm。结果 野马追内酯A在0.02~1.88 μg,野马追内酯B在0.44~4.44 μg内与峰面积呈良好的线性关系。金丝桃苷在0.101~1.414 μg内与峰面积呈良好的线性关系。结论 本实验方法准确、快速、灵敏、重复性好,在该色谱条件及样品处理方法下,可以准确测定野马追中野马追内酯A,B和金丝桃苷的含量。

Determination of Eupalinolide A, B and Hyperin in Eupatorium Lindleyanum DC by HPLC

Abstract:
OBJECTIVE To establish a method for the determination of the content of eupalinolide A, B and hyperin in Eupatorium lindleyanum DC from 6 different areas by HPLC. METHODS For eupalinolide A, B, Agilent Extend-C18 column (4.6 mm×150 mm, 5 μm) was used under the mobile phase of acetonitrile-water(27∶73) at the flow rate of 1.0 mL·min-1. The detection wavelength was 220 nm and the column temperature was maintained at 30 ℃. For hyperin, SB-C18 column was used under the mobile phase of acetonitrile-1% Acetic acid(12∶88) at a flow of 1.0 mL·min-1. The detection wavelength was 255 nm and the column temperature was maintained at 30 ℃. RESULTS The standard curve of eupalinolide A was linear in the range of 0.02-1.88 μg and eupalinolide B was linear in the range of 0.44-4.44 μg. The standard curve of hyperin was linear in the range of 0.101-1.414 μg. CONCLUSION This method was simple, accurate and reproducible for the determination of eupalinolide A, B and hyperin in Eupatorium lindleyanum DC.
