摘要: |
目的 建立高效液相色谱法研究大鼠口服西河柳总黄酮后柽柳素和山奈甲黄素的组织分布。方法 采用Waters C18色谱柱(4.6 mm×250 mm,5 μm),以甲醇-水(pH 2.7)为流动相,流速1.0 mL·min-1,检测波长254 nm,柱温35 ℃,组织样品用固相萃取法处理。结果 在多种组织样品中同时检测到柽柳素和山奈甲黄素,柽柳素在肺、心中分布最高,山奈甲黄素最高分布在心,次之为肺。结论 利用高效液相色谱法可以同时进行柽柳素和山奈甲黄素组织分布研究。 |

OBJECTIVE To develop an HPLC method for simultaneous determination of tamarixetin and kaempferide in rat tissues after oral administration of the total flavonoides from Tamarix chinensis (TFT). METHODS Using Waters C18 column (4.6 mm×250 mm, 5 μm), the mobile phase was methanol-0.15% aqueous formic acid solution (pH 2.7) at a flow rate of 1.0 mL·min-1 and detection wavelength was 254 nm, the column temperature was kept at 35 ℃. RESULTS Tamarixetin and kaempferide were simultaneous determined in several tissues, the highest level of tamarixetin was observed in lung, then in heart; and the highest concentration of kaempferide was determined in heart, then in lung. CONCLUSION The HPLC method is good in the aspect of easy operation and veracity, and can be used to study tissues distribution of tamarixetin and kaempferide. |
