目的 建立微乳液相色谱法同时测定广东紫珠中5,4′-二羟基-3,7,3′-三甲氧基黄酮、鼠李秦素、华良姜素、岳桦素、毡毛美洲茶素5种黄酮成分。方法 微乳体系组成的流动相为:2.2%SDS -1.5%正辛烷-7.75%正丁醇- 0.5%三乙胺-88%水(用H3PO4调节pH 3.7),色谱柱为Hypersil BDS C18(4.6 mm×150 mm,5 μm)柱;检测波长350 nm;流速0.8 mL·min-1;进样量10 μL;柱温为30 ℃。结果 广东紫珠中5种黄酮成分在30 min内达到完全分离,在1.520~72.25 μg·mL-1内,5个黄酮成分的线性相关系数r≥0.999 5,平均回收率99.6%~100.5%。结论 本方法可应用于中广东紫珠中5种黄酮成分的含量分析。 |

OBJECTIVE To establish an efficient microemulsion liquid chromatography(MELC) method and validate for simultaneous determination of five flavonoids (5,4′-dihydroxy-3,7,3′-trimethoxyflavone, rhamnatin, kumatakenin, ermanine and velutin) in Callicarpa kwangtungensis Chun.. METHODS The MELC separation was performed on a Hypersil BDS Cl8 column(4.6 mm×150 mm, 5 μm) with the mixture of 2.2% SDS-1.5% octane-7.75% n-butanol-0.5% triethylamine-88% water, the pH was adjusted to 3.7 with phosphoric acid as the mobile phase. The flow rate was set at 0.8 mL·min-1 and the eluent was detected at 350 nm for the five flavonoids.RESULTS The calibration curves of the five flavonoids were linear (r≥0.999 5) over the concentration range of 1.520-72.25 μg·mL-1. The mean recoveries were 99.6%-100.5%. CONCLUSION The results indicate that the optimized method is successfully applied to the analysis of five important flavonoids in Callicarpa kwangtungensis Chun.. |
