摘要: |
目的 研究灵芝三萜(ganoderma triterpene,GT)对小鼠非酒精性脂肪肝的治疗作用。方法 以复方高脂饲料连续喂养小鼠5周,建立非酒精性脂肪肝小鼠模型。除正常对照组的8只小鼠外,将造模成功小鼠40只随机分为5组,分别为模型对照组、阳性药物组(壳脂胶囊100 mg·kg-1,1次·d-1)、GT高、中、低(200,100,50 mg·kg-1,1次·d-1)剂量组。连续给药6周后,检测血清和肝脏中生化指标,做肝脏组织病理学检查。结果 非酒精性脂肪肝小鼠经GT治疗后,GT高、中剂量组都表现出降低非酒精性脂肪肝小鼠血清中总胆固醇(TC)、甘油三脂(TG)含量和天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)活性的作用,降低非酒精性脂肪肝小鼠肝脏中TG、TC、丙二醛(MDA)含量,升高超氧化物岐化酶(SOD)活力的作用(P<0.05或P<0.01);组织病理学检查发现脂肪肝程度明显减轻,肝细胞坏死程度减轻。结论 GT对高脂饲料所致非酒精性脂肪肝小鼠具有保肝调脂作用。 |

OBJECTIVE To investigate the therapeutic effect of ganoderma triterpene(GT) on non-alcoholic fatty liver in mice. METHODS A model of the non-alcoholic fatty liver was constructed by feeding mice with high glucose and fatty diet for 5 weeks. Forty mice models with non-alcoholic fatty liver were randomly divided into five groups, model groups, Kezhi capsule groups(100 mg·kg-1), GT groups (200, 100, 50 mg·kg-1). The therapeutic effects of GT on non-alcoholic fatty liver and histopathological changes of mice liver were observed. RESULTS Compared with the non-alcoholic fatty liver model group, after treatment with GT, the mice serum levels of total cholesterol(TC), triglyceride(TG), alanine transaminase(ALT) and aspartate transaminase(AST) were significantly decreased(P<0.05 or P<0.01). The hepatic tissue contents of TC, TG and malondialdehyde(MDA) also significantly decreased (P<0.05 or P<0.01). The superoxide dismutase(SOD) activity was higher than that of mice in model group (P<0.05 or P<0.01). And the pathobiology of liver dramatically showed the repairing of hepatic steatosis. CONCLUSION GT has the hypolipideic function with liver protection on high fat diet-induced non-alcoholic fatty liver disease in mice. |
