目的 本综述简要介绍先导化合物优化阶段,HPLC-MS/MS应用于药物代谢和药动学(DMPK)中筛选一系列新的化学实体。方法 查阅有关文献,进行分析总结。结果 HPLC-MS/MS在支持体内或体外试验以及定性分析,如代谢物鉴定等方面均有广泛应用。结论 液质联用已成为DMPK筛选强有力且必不可少的分析手段。 |

Abstract: |
OBJECTIVE This review summarizes application of high performance liquid chromatography coupled to a tandem mass spectrometer(HPLC-MS/MS) methodology in drug metabolism and pharmacokinetic screens for a series of new chemical entities during the lead optimization stages. METHODS Relevant literatures were consulted, analysis and summary were made. RESULTS The HPLC-MS/MS methods were widely used to support both in vitro and in vivo experiments and for the qualitative assays, such as metabolite identification. CONCLUSION HPLC-MS/MS is a powerful and indispensable analytical tool in drug metabolism and pharmacokinetic screening. |
