摘要: |
目的 探讨醉香含笑树皮提取物(extrat of barks of Michelia macclurei Dandy.,EBMMD)诱导人肝癌细胞HepG2凋亡的作用及相关机理。方法 采用倒置生物显微镜、流式细胞仪、Western-blotting等技术,考察EBMMD对HepG2细胞凋亡及COX-2蛋白表达的影响。结果 流式细胞术分析可见,EBMMD能明显诱导HepG2细胞凋亡,且呈浓度依赖关系;倒置生物显微镜观察可见细胞凋亡的形态学改变;随EBMMD浓度增高,细胞内COX-2蛋白的表达逐渐减少,且当EBMMD浓度达25.00 μg·mL-1时,能明显抑制COX-2蛋白表达。结论 EBMMD可以明显诱导HepG2细胞凋亡,其作用机理可能与下调细胞内COX-2蛋白表达进而诱导其凋亡有关。 |

OBJECTIVE To explore the effect and possible mechanism of the extract of barks of Michelia macclurei Dandy.(EBMMD) on apoptosis-inducing in HepG2 cell lines in vitro. METHODS The effect of apoptosis-inducing of EBMMD on HepG2 cell lines and expression of COX-2 protein were explored by inverted microscope and flow cytometry assay as well as Western-blotting assay. RESULTS The apoptosis-inducing of EBMMD on HepG2 cells was observed in concerntration-dependent manner by flow cytometry, and the cell morphology change was observed by inverted microscope. EBMMD showed growing inhibitory effects on the expressions of COX-2 protein in concerntration-dependent manner and significant inhibitory effect on the expression of COX-2 protein at a high concentration(25.00 μg·mL-1) . CONCLUSION EBMMD can significantly induce HepG2 cells apoptosis, the possible mechanism may relate to downregulate the expression of COX-2 protein to induce apoptosis. |
