HPLC同时测定浙江蜡梅中3种黄酮类成分的含量

摘要:
目的 建立同时测定浙江蜡梅中3种黄酮类成分含量的高效液相色谱法。方法 采用Agilent Zorbax SB-C18色谱柱(4.6 mm×250 mm,5 μm),柱温为30 ℃;流动相为乙腈-0.1%磷酸溶液,梯度洗脱,流速为1.0 mL·min-1;检测波长为360 nm。结果 芦丁、槲皮素和山奈酚的线性范围分别为4.057~507.2,0.686 1~171.5,0.216 9~81.35 μg·mL-1,r分别为0.999 3,0.999 6,0.999 9;平均加样回收率分别为99.97%(RSD=0.97%),100.13%(RSD=1.06%),99.35%(RSD=1.39%)。结论 该方法同时测定浙江蜡梅中的芦丁、槲皮素和山奈酚的含量,简便易行、快速准确、重复性好,可用于浙江蜡梅的质量控制。

Abstract:
OBJECTIVE To establish an HPLC method to simultaneously determine three kinds of flavonoids in Chimonanthus zhejiangensis M. C. Liu. METHODS The Agilent Zorbax SB-C18 chromatographic column (4.6 mm×250 mm, 5 μm) was adopted. The column temperature of was 30 ℃. The mobile phase was acetonitrile-0.1% phosphoric acid solution with the gradient elution at the flow rate of 1.0 mL·min-1. The detection wavelength was 360 nm. RESULTS Good linear correlation of rutin, quercetin and kaempferol were found in the ranges of 4.057-507.2, 0.686 1-171.5, 0.216 9-81.35 μg·mL-1(r=0.999 3, 0.999 6, 0.999 9), respectively. The average recoveries were 99.97%(RSD=0.97%), 100.13%(RSD=1.06%), 99.35%(RSD=1.39%). CONCLUSION This method is simple, accurate and repeatable for simultaneously determination of the contents of rutin, quercetin and kaempferol in Chimonanthus zhejiangensis M. C. Liu, and applicable for the quality control of Chimonanthus zhejiangensis M. C. Liu.
