目的 建立同时测定复方红花喷雾剂中羟基红花黄色素A(hydroxysafflor yellow A,HSYA)与绿原酸含量的方法。方法 采用双波长HPLC,色谱柱:Shim-pack VP-ODS(250 mm×4.6 mm,5 μm),流动相:乙腈-0.4%磷酸溶液(12∶88),流速为1.0 mL·min-1,检测波长:403 nm(HSYA)和327 nm(绿原酸),柱温为30 ℃。结果 HSYA、绿原酸的浓度分别在0.612~12.24,0.632~12.64 μg·mL-1内与各自峰面积呈良好的线性关系(r均为0.999 9);平均加样回收率分别为98.34%和99.82%,RSD分别为1.16%和1.64%(n=9)。结论 该方法操作简便、快速,结果可靠,具有良好的重复性,可用于复方红花喷雾剂中HSYA与绿原酸的含量测定。 |

OBJECTIVE To establish a method for the simultaneous determination of hydroxysaffor yellow A(HSYA) and chlorogenic acid in Fufang Honghua spray. METHODS The dual-wavelength HPLC was adopted. The Shim-pack VP-ODS column(250 mm×4.6 mm, 5 μm) was used with mobile phase of acetonitrile-0.4% phosphonic acid(12∶88) at flow rate of 1.0 mL·min-1, and the detection wavelength was set at 403 nm for HSYA and 327 nm for chlorogenic acid. The column temperature was 30 ℃. RESULTS The method displayed good linearity over the concentration ranges of 0.612-12.24 μg·mL-1 for HSYA and 0.632-12.64 μg·mL-1 for chlorogenic acid(r=0.999 9), respectively. The average recoveries were 98.34% and 99.82%, with the RSD of 1.16% and 1.64% (n=9), respectively. CONCLUSION This method is simple, rapid, reliable and reproducible. It can be used for determination of HSYA and chlorogenic acid in Fufang Honghua spray. |
