HPLC同时测定大血藤中绿原酸、咖啡酸和香草酸的含量

摘要:
目的 建立同时测定大血藤各部位绿原酸、咖啡酸和香草酸含量的高效液相色谱方法。方法 色谱柱为Kromasil C18柱(4.6 mm×250 mm,4.5 μm),流动相:A为1%冰醋酸,B为甲醇,以A∶B=68∶32等度洗脱;柱温:30 ℃;流速:1 mL·min-1;检测波长:327 nm;进样量:20 μL。结果 在测定样品中,绿原酸和咖啡酸在乙酸乙酯部位含量最高,分别为4.52%,0.11%,香草酸在二氯甲烷部位含量最高,为5.8%。结论 该方法简便、准确、重复性高,可用于大血藤中绿原酸、咖啡酸、香草酸含量的测定。

Abstract:
OBJECTIVE To develop an HPLC method for simultaneous determination of chlorogenic acid, caffeic acid and vanillic acid in each fraction of Sargentodoxae Caulis. METHODS The analysis was carried on a column of Kromasil C18(4.6 mm×250 mm, 4.5 μm). Isocratic elution, mobile phase consisted of solvent A (1%, solution of HAc in water) and solvent B (methanol) at the ratio of 68∶32, the flow rate was 1.0 mL.min-1, the injection volume was 20 μL, the detection wavelength was 327 nm, the column temperature was at 30 ℃. RESULTS Ethyl acetate fraction had the highest concentration of chlorogenic acid (4.52%) and caffeic acid (0.11%) and dichloromethane fraction possessed the highest concentration of vanillic acid of 5.8%. CONCLUSION This method is simple, accurate, rapid and suitable for the usual quality control of Sargentodoxae Caulis.
