摘要: |
目的 基于NF-κB通路探讨延龄草醇提取物对CCl4所致大鼠急性肝损伤的保护作用及机制。方法 把48只SD大鼠,随机分为空白对照组、模型组、联苯双酯组(200 mg·kg-1)、延龄草醇提取物低、中、高剂量组(100,500,1 000 mg·kg-1)6组。以CCl4诱导急性肝损伤模型,造模后灌胃给药,治疗持续一段时间后取材。称重后计算大鼠肝脏脏器系数;检测各组大鼠血清中ALT、AST的活性;利用HE染色切片观察各组肝组织形态学的变化;以RT-PCR法检测各组大鼠肝组织中TNF-α、IL-1β,IL-6的基因表达;通过western blot检测各组肝组织中NF-κB的表达量。结果 与模型组相比,延龄草醇提物各治疗组可降低肝脏脏器系数,高剂量组肝小叶结构完整,肝细胞排列规则,大小均匀;RT-PCR结果显示,延龄草醇提取物能够显著降低TNF-α、IL-1β、IL-6的表达。western blot结果显示,延龄草醇提物显著地降低了NF-κB的表达。结论 延龄草醇提取物能够通过抑制NF-κB的表达,保护CCl4诱导的大鼠急性肝损伤。 |

OBJECTIVE To explore the protective effect of Trillium tschonoskii Maxim.(TTM) based on NF-κB signaling pathway. METHODS The 48 male SD rats were randomly divided into control group, model group, bifendate group (200 mg·kg-1), TTM low, middle and high dose group (100, 500, 1 000 mg·kg-1). After the establishment of liver injury model induced by CCl4, the rats was treated for some time, then killed for samples. The rat liver was weighed to calculate the liver coefficient; serum ALT and AST were detected; the liver tissue morphology were observed by HE staining; RT-PCR was used to detecte the expression of TNF-α, IL-1β, IL-6; Western blot was used to detecte the protein expression of NF-κB. RESULTS compared with the model group, the liver coefficient was reduced by TTM, especially in the high dose group, the morphological structure of hepatic lobule was complete, liver cells arranged with regular size; RT-PCR showed that TTM alcohol extract could decrease gene expression of TNF-α, IL-1β, IL-6 significantly, and Western blot also showed that TTM could extract decease the protien expression of NF-κB compared with model group. CONCLUSION TTM improves the acute liver injury induced by CCl4 in rats, the mechanism may be related to inhibiting the expression of NF-κB. |
