基于HPLC特征图谱和主要成分含量测定的龙胆药材干燥方法研究

摘要:
目的 通过测定龙胆药材不同干燥方法所得样品的HPLC特征图谱及3种主要成分的含量,确定龙胆药材的最佳干燥方法。方法 采用Welchrom C18色谱柱(4.6 mm×250 mm,5 μm);流动相:甲醇-水,梯度洗脱;流速:1.0 mL·min-1;进样量:10 μL;检测波长:240 nm;柱温:25℃。结果 建立了龙胆药材鲜品及不同方法干燥后样品的HPLC特征图谱,同时测定了样品中3种主要成分的含量。结论 龙胆药材的最佳干燥方法为阴干法。

Abstract:
OBJECTIVE To confirm the optimal drying method of Gentiana scabra Bge. based on HPLC characteristic fingerprint and contents determination of three main components in the samples. METHODS The condition was as followes:Welchrom C18 column(4.6 mm×250 mm,5 μm); mobile phase:methanol-water with gradient elution; flow rate:1 mL·min-1; injection volume:10 μL; detection wavelength:240 nm; column temperature:25℃. RESULTS The HPLC characteristic fingerprint of Gentiana scabra Bge. was established, while the contents of 3 main components in samples of fresh and different drying methods were determined. CONCLUSION The shade drying is the optimal drying method of Gentiana scabra Bge.
