人参超微粉对正常和免疫力低下小鼠免疫功能的影响

摘要:
目的 探讨人参超微粉化后是否仍具有增强免疫的作用。方法 实验主要分为2个部分,第一部分:将96只Balb/c小鼠随机分成2个免疫组(免疫Ⅰ组、免疫Ⅱ组),每个免疫组按体质量随机分为空白组、人参超微粉3个剂量组(166.7,333.3,500.0 mg·kg-1)。灌胃32 d,免疫Ⅰ组进行脾淋巴细胞增殖实验和NK细胞活性实验,测定脏器(脾、胸腺)指数、刺激指数和NK细胞活性;免疫Ⅱ组进行迟发型超敏反应(DTH)和血清溶血素实验,测定小鼠足跖肿胀度和抗体积数;第二部分:将60只ICR小鼠按体质量随机分为空白组、模型组、人参超微粉3个剂量组(166.7,333.3,500.0 mg·kg-1),连续灌胃13 d,于第5天开始,除空白组外,连续3 d腹腔注射环磷酰胺40 mg·kg-1,建立免疫力低下模型。测定其白细胞数目,脏器(脾、胸腺)指数,血清IL-2、IL-4、IgG、IgM水平及脾匀浆LDH、ACP活性。结果 第一部分:人参超微粉3个剂量组小鼠胸腺指数升高;人参超微粉333.3,500.0 mg·kg-1组小鼠抗体积数及注射绵羊红细胞24 h后足跖肿胀度显著高于空白组,同时能增强NK细胞活性。第二部分:人参超微粉333.3,500.0 mg·kg-1组小鼠白细胞数目明显增加,血清中IL-2、IL-4、IgG、IgM水平升高及脾匀浆LDH、ACP活性增强。结论 一定剂量的人参超微粉对正常和免疫力低下小鼠具有免疫调节作用,提示将人参超微粉化后仍具有增强免疫的作用,这为人参超微粉的进一步开发利用提供了实验依据。

Effects of Superfine Powder of Ginseng on Immune Functions in Normal and Immunocompromised Mice

Abstract:
OBJECTIVE To investigate if ginseng has the effect on enhancing the immunity after micronizing. METHODS The experiment was divided into two parts, part one:96 Balb/c mice were randomly divided into two immunized groups (immune groupⅠand immune groupⅡ). According to body weight, each immunized group was randomly divided into 4 groups:blank group, three dose groups of superfine powder of ginseng (SPG) (166.7, 333.3, 500.0 mg·kg-1). After gavaged for 32 d, immune groupⅠ:organ index (spleen and thymus), SI (Con A), NK cell activities were measured by splenic lymphocyte proliferation ability and NK cell activities test. Immune groupⅡ:the degree of plantar swelling in mice before and after SRBC stimulation and the anti-volume level in serum was measured by DTH and serum hemolysin experiment. Part two:according to body weight, 60 ICR mice was randomly divided into 5 groups:blank group, model group, three dose groups of SPG (166.7, 333.3, 500.0 mg·kg-1), each group were administrated for 13 d. Start from the 5th day of the experiment, each group was injected CTX 40 mg·kg-1 intraperitoneally for 3 d expect blank group to establish immunocompromised model. The number of WBC, organ (spleen and thymus) index, the levels of IL-2, IL-4, IgG and IgM in serum and the activities of LDH and ACP in spleen homogenate were measured. RESULTS Part one:spleen and thymus index in each groups of SPG and the NK cell activities of SPG 333.3, 500.0 mg·kg-1 group was significantly increased. The antibody volume and the degree of plantar swelling before and after SRBC stimulation in SPG 333.3, 500.0 mg·kg-1 group were higher than the blank group. Part two:The number of WBC, the levels of IL-2, IL-4, IgG and IgM in serum and the activities of LDH, ACP in spleen homogenate were significantly increased in SPG 333.3, 500.0 mg·kg-1 group. CONCLUSION A certain dose of SPG has the effect on enhancing immunity in normal and immunocompromised mice, suggesting that ginseng still has the effect on enhancing the immunity after micronizing, which provides the experimental basis for the further development and utilization of SPG.
