柱前衍生-HPLC测定不同等级的梅花鹿鹿茸饮片中氨基酸的含量

摘要:
目的 建立柱前衍生-HPLC测定不同等级梅花鹿鹿茸饮片中17种氨基酸成分含量的方法,并比较氨基酸含量的差异。方法 采用HPLC,以异硫氰酸苯酯进行柱前衍生,梯度洗脱的方法,检测不同等级梅花鹿鹿茸饮片中17种氨基酸的含量。结果 不同等级的梅花鹿鹿茸饮片的总氨基酸成分含量差异显著。氨基酸含量蜡片 > 粉片 > 纱片 > 骨片。结论 梅花鹿鹿茸饮片的质量受采割部位的影响。该方法适用于梅花鹿鹿茸饮片中氨基酸成分的含量测定,可为其等级的制定提供理论依据。

Determination of Amino Acids in Different Grades of Antler Pieces from Cervus Nippon by Pre-column Derivatization-HPLC

Abstract:
OBJECTIVE To establish a method for the determination of 17 amino acids contents in different grades of antler pieces from Cervus nippon using pre-column derivatization-HPLC, and to compare the difference of amino acid contents. METHODS The contents of 17 kinds of amino acids in different grades of antler pieces were detected by HPLC with pre-column derivatization and gradient elution with phenyl isothiocyanate. RESULTS There were significant differences in the total amino acid contents of different grades of antler pieces. Amino acid contents from high to low was wax, powder, sand and bone chips. CONCLUSION The quality of antler pieces is affected by the position of harvest. The method is suitable for the determination of the amino acid contents in the antler pieces, which can provide a theoretical basis for the formulation of antler pieces.
