R语言计算药动学参数及生物等效性分析

摘要:
目的 使用R语言计算非房室模型药动学参数,并进行生物等效性分析。方法 使用R语言中"NonCompart"包进行非房室模型药动学参数的计算。使用"nlme"包建立线性混合效应模型,进行多因素方差分析。使用"PowerTOST"包计算90%置信区间。将结果与Phoenix WinNonlin 6.4软件的计算结果对比。结果 联合使用R包进行非房室模型药动学参数的计算与生物等效性分析的结果与Phoenix WinNonlin 6.4软件计算结果一致。结论 可以通过R语言实现非房室模型药动学参数的计算与生物等效性分析,且结果可靠。

Abstract:
OBJECTIVE To calculate the pharmacokinetic parameters of non-compartmental models and analysis bioequivalence by use R software. METHODS The pharmacokinetic parameters of the non-compartmental model were calculated by the "NonCompart" package in the R software. A linear mixed-effects model was built using the "nlme" package for multivariate analysis of variance. Use the "PowerTOST" package to calculate the 90% confidence interval of geometric mean ratio. The results was compared with the Phoenix WinNonlin 6.4 software. RESULTS The results of pharmacokinetic parameters of the non-compartmental model and bioequivalence analysis by using the R software were consistent with result which calculated by Phoenix WinNonlin 6.4 software. CONCLUSION The pharmacokinetic parameters of non-compartmental model calculation and bioequivalence analysis can be achieved by R software and the result is reliable.
