摘要: |
目的 建立陈皮配方颗粒的UPLC指纹图谱,为该药的整体质量评价提供依据。方法 选用Waters Acquity UPLC BEH C18色谱柱(2.1 mm×10 mm,1.7 μm),以乙腈-0.2%磷酸水溶液为流动相,梯度洗脱,流速0.3 mL·min-1,检测波长283 nm,进样体积2 μL。以橙皮苷为参照,绘制12批样品的UPLC图谱,采用"中药色谱指纹图谱相似度评价系统"(2012年版)进行相似度评价,确定共有峰,并采用SPSS Statistics 24软件进行聚类分析和主成分分析。结果 12批样品的UPLC图谱有18个共有峰,相似度均>0.970,表明12批样品的化学成分一致性好。12批样品可聚为2类,S1,S2,S3,S4,S5,S6聚为一类,S7,S8,S9,S10,S11,S12聚为一类。经主成分分析,2个主成分因子的累积方差贡献率为89.424%,S2样品的主成分因子综合得分最高,整体质量最好。结论 所建UPLC指纹图谱及聚类分析和主成分分析结果可为陈皮配方颗粒的质量评价提供参考。 |

OBJECTIVE To establish an UPLC fingerprint method of Citri Reticulatae Pericarpium formula granules to provide a reference for evaluation of the overall quality. METHODS UPLC was performed on a Waters Acquity UPLC BEH C18 analytical column(2.1 mm×10 mm, 1.7 μm). Acetonitrile and 0.2% phosphoric acid was adopted as the mobile phase by gradient elution at the flow rate of 0.3 mL·min-1. The detector wavelength was set at 283 nm and the injection volume was 2 μL. Using hesperidin as reference, UPLC fingerprints of 12 batches of Citri Reticulatae Pericarpium formula granules were determined. The similarity of 12 batches of samples was evaluated by TCM Chromatographic Fingerprint Similarity Evaluation System (2012 edition) to confirm common peak. Cluster analysis and principal component analysis were performed by using SPSS Statistics 24 software. RESULTS There were 18 common peaks in UPLC chromatograms of 12 batches of samples, the similarity of which was >0.970, indicating the chemical composition of 12 batches of samples were consistent. Twelve batches of samples were clustered into 2 categories; S1, S2, S3, S4, S5 and S6 were clustered into one category; S7, S8, S9, S10, S11 and S12 were clustered into the other category. By principal component analysis, the accumulative contribution rate of two main component was 89.424%, and comprehensive score of S2 was the highest with the best quality. CONCLUSION Established fingerprint, the results of cluster analysis and principal component analysis can provide reference for quality evaluation of Citri Reticulatae Pericarpium formula granules. |
