葛仙米多糖对白色念珠菌诱导的免疫低下小鼠免疫功能的影响

摘要:
目的 考察葛仙米多糖对免疫低下小鼠免疫功能的影响。方法 BALB/c小鼠分成对照组,模型组,葛仙米多糖低、中、高剂量(0.1,0.2,0.4 g·kg-1)组,阳性药组(玉屏风颗粒2.5 g·kg-1),动物分组后持续灌胃给药28 d,第29天除对照组外,其余组小鼠尾静脉注射白色念珠菌每只5×104 CFU,造成免疫低下模型。注射白色念珠菌后第14天,解剖,检测小鼠体质量、脏器指数、血常规、脾组织形态、脾淋巴细胞增殖能力、腹腔巨噬细胞吞噬能力和凋亡以及Bcl-2和Bax蛋白的表达。结果 与对照组比较,模型组小鼠体质量下降,白细胞下降,中性粒细胞增加,胸腺、脾脏指数降低,脾脏组织形态损伤,脾淋巴细胞增殖能力降低,腹腔巨噬细胞吞噬能力降低,腹腔巨噬细胞凋亡增加,Bcl-2/Bax蛋白比值降低,P均<0.01。与模型组比较,葛仙米多糖各剂量组脏器指数显著增大(P<0.01),中性粒细胞显著减少(P<0.01),脾组织形态修复,脾淋巴细胞增殖能力明显增加(P<0.05或P<0.01),腹腔巨噬细胞的吞噬能力显著改善(P<0.01),腹腔巨噬细胞的凋亡显著降低(P<0.01),Bcl-2/Bax蛋白比值显著升高(P<0.01)。结论 葛仙米多糖能显著增强免疫低下小鼠的免疫功能。

Abstract:
OBJECTIVE To investigate the effects of Nostoc sphaeroides Küte polysaccharide on immune function of the immunodeficiency mice. METHODS BALB/c mice were divided into control group, model group, low, medium and high-dose of Nostoc sphaeroides Küte polysaccharide groups(0.1,0.2,0.4 g·kg-1) and positive drug group(Yupingfeng granules 2.5 g·kg-1). Animals were given intragastric administration for 28 d. On the 29th day, the rats in other groups were injected with Candida albicans(5×104 CFU) in the tail vein except for the control group, which resulted in an immunodeficiency model. Observations of the weight, organ index, blood routine, spleen morphology, spleen lymphocyte proliferation, peritoneal macrophage phagocytosis and apoptosis, Bcl-2 and Bax protein expression were carried out on the 14th day after injection of Candida albicans. RESULTS Compared with the control group, the rats in model group lost weight, while white blood cells was decreased, neutrophilic granulocytes was increased, thymus and spleen index were decreased, spleen morphology was injured, the proliferation of spleen lymphocytes was decreased, the phagocytic ability of macrophages was decreased, the apoptosis of peritoneal macrophages was increased, Bcl-2/Bax was decreased, all P<0.01. Compared with the model group, the thymus index and the spleen index of rats in all dose groups of Nostoc sphaeroides Küte polysaccharide was increased significantly(P<0.01), while neutrophilic granulocytes was decreased(P<0.01), the spleen morphology was repaired, the proliferation of spleen lymphocytes was increased significantly(P<0.05 or P<0.01), the phagocytosis of peritoneal macrophages was increased significantly(P<0.01), the apoptosis of peritoneal macrophages was decreased significantly(P<0.01), Bcl-2/Bax was increased (P<0.01). CONCLUSION Nostoc sphaeroides Küte polysaccharide can significantly enhance the immune function of immunocompromised mice.
