摘要: |
目的 建立同时测定川芎清脑颗粒中绿原酸、阿魏酸、升麻素苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、蛇床子素的HPLC方法。方法 采用Agilent Poroshell 120 EC-C18色谱柱(150 mm×4.6 mm,2.7 μm),流动相为甲醇-0.05%的磷酸溶液,梯度洗脱,检测波长分别为330 nm(绿原酸、阿魏酸、蛇床子素),280 nm(黄芩苷、汉黄芩苷、黄芩素、汉黄芩素),254 nm(升麻素苷)。结果 绿原酸、阿魏酸、升麻素苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、蛇床子素的线性范围分别为4.97~37.29,5.29~39.68,2.54~19.08,35.46~265.94,25.65~192.37,2.40~18.02,1.66~12.47,2.64~19.80 μg·mL-1;精密度、稳定性、重复性试验的RSD<2%;平均回收率(n=6)均>90%。结论 该方法精确可信、操作简便快捷,可用于川芎清脑颗粒中各成分同时测定。 |

Abstract: |
OBJECTIVE To establish an HPLC method for the simultaneous determination of chlorogenic acid, ferulic acid, prim-O-glucosylcimifugin, baicalin, wogonoside, baicalein, wogonin and osthole in Chuanxiong Qingnao granules. METHODS The column was Agilent Poroshell 120 EC-C18(150 mm×4.6 mm, 2.7 μm) with mobile phase of methanol-0.05% phosphoric acid(gradient elution), the detection wavelength was 330 nm(for chlorogenic acid, ferulic acid, osthole), 280 nm(for baicali, wogonoside, baicalein, wogonin) and 254 nm(for prim-O-glucosylcimifugin). RESULTS The linear range was 4.97-37.29 μg·mL-1 for chlorogenic acid, 5.29-39.68 μg·mL-1 for ferulic acid, 2.54-19.08 μg·mL-1 for prim-O-glucosylcimifugin, 35.46-265.94 μg·mL-1 for baicalin, 25.65-192.37 μg·mL-1 for wogonoside, 2.40-18.02 μg·mL-1 for baicalein, 1.66-12.47 μg·mL-1 for wogonin, 2.64-19.80 μg·mL-1 for osthole, respectively; RSDs of precision, stability and reproducibility tests were < 2.0%; The average recovery in sample(n=6) was > 90%. CONCLUSION The method is accurate, reliable, simple and fast, and can be used for simultaneous determination of multiple components in Chuanxiong Qingnao granules. |
