柿叶总黄酮对高脂血症大鼠脂肪肝和脂蛋白代谢相关酶的影响

摘要:
目的 研究柿叶总黄酮(PLF)对饮食性高脂血症大鼠肝脏脂肪变性的影响。方法 建立饮食性高脂血症大鼠模型,将高脂血症大鼠按照血清TC水平随机分成5组(每组10只),分别为:高脂模型组、PLF高剂量组、PLF中剂量组、PLF低剂量组和洛伐他汀组。连续给药4周后检测各组大鼠天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、脂蛋白酯酶(LPL)、肝酯酶(HL)和血脂水平等各项指标,并用HE染色法观察大鼠肝组织病理形态学改变情况。结果 与模型组比较,各用药组TC,TG,LDL-C,AST,ALT水平降低,HDL-C,LPL,HL水平升高,均具有统计学差异(P<0.01或P<0.05);各给药组肝重、肝脏系数显著降低(P<0.01或P<0.05),肝组织脂肪变性程度均有不同程度的改善。结论 PLF对高脂血症大鼠的脂质代谢紊乱有显著的调节作用,对防治高脂性脂肪肝有良好的作用。

Abstract:
OBJECTIVE To explore the effect of persimmon leaf total flavonid (PLF) on the hyperlipidemic fatty liver and enzyme of lipoprotein metabolism in rats. METHODS Fifty healthy SD rats were raised by hyper-fatty animal feed for 6 weeks to establish the hyperlipidemia rats model. Then the hyperlipidemia rats were divided into 5 groups randomly according to the serum TC: model group, high dose group of PLF, medium dose group of PLF, low dose group of PLF, and lovastatin group. Serum lipoprotein level, aspartate aminotransferase(AST), alanine aminotransferase(ALT), lipoprotein lipase(LPL) and hepatic lipase(HL) were tested after treated with corresponding drugs once per day for 4 weeks, in addition to liver tissue physiological form were observed by HE dye way. RESULTS Compared with the model group, the levels of the serum TC, TG, LDL-C, AST and ALT were decreased significantly, while HDL-C, LPL and HL were increased significantly (P<0.05 or P<0.01). In all treated groups, the liver weight, liver coefficient were decreased significantly (P<0.05 or P<0.01), and the liver adipose degeneration were ameliorated in some degree after the treatment with drugs. CONCLUSION PLF have marked benefit effects on prevention and treatment of the hyperlipemia and liver adipose degeneration.
