大孔树脂分离纯化延胡索总生物碱的工艺研究

摘要:
目的研究大孔树脂分离纯化延胡索总生物碱的工艺条件及参数。方法以延胡索总生物碱提取率为评价指标筛选大孔树脂。结果经试验表明分离纯化的最佳工艺为D-101大孔树脂,上样量为延胡索药材与树脂体积比为1∶1(g·mL-1);洗脱速度0.5 BV·h-1;去离子水洗脱杂质,洗脱体积为6 BV,80%乙醇洗脱总生物碱,洗脱体积为6 BV。结论该工艺简单可行,分离效果好,能满足于大生产的要求。

Abstract:
OBJECTIVE To explore the purification method of gross alkaloid in Rhizoma Corydalis with macropopous adsorbing resin.METHODS The extracting yield of the gross alkaloid were taken as index to select the macroporous resin.RESULTS Optium conditions for isolation and purification were indentified, I.e. D-101 type macropopous adsorbing resin,adsorbing rate was 0.5 BV·h-1,water volume for removing impurities was 6 BV,concentration of the ethanol was 80%,and volume of ethanol was 6 BV.CONCLUSION The method is simple and feasible,with good separating effect which can meet industrial requirements.
