心脏疾病治疗药物的临床疗效提升严重受限于药物的非特异性分布。本综述系统评述了心脏靶向药物递送系统,重点围绕仿生心脏天然修复途径、识别心肌生物标志、借助病灶血管EPR效应、响应病灶特殊微环境等智能载体设计策略。此外,本综述还探讨了局部给药方式与多种心脏靶向策略联合应用,以促进药物跨生理屏障的心脏病灶浓集给药。基于此,本综述深入剖析了心脏靶向递送系统面临的挑战,并展望了其临床转化前景。通过系统阐述心脏靶向策略的智能设计和应用前景,旨在为心脏靶向递药的研究发展提供重要参考。 |

Abstract: |
Clinical efficacy of therapeutic drugs for cardiovascular diseases is severely limited by the nonspecific distribution of the drugs. The present review systematically summarizes cardiac-targeted drug delivery systems, focusing on intelligent design strategies that mimic endogenous cardiac repair pathways, recognize myocardial biomarkers, leverage the EPR effect of lesion vessels, and respond to the unique microenvironment of lesions. Moreover, the review discusses local drug administration methods and combinative strategies of various cardiac targeting to promote concentrated drug delivery at cardiac lesion sites across physiological barriers. Accordingly, the present review provides an in-depth analysis of the challenges faced by cardiac-targeted delivery systems and looks forward to their clinical transformation prospects. Reviewing the intelligent design and application prospects of cardiac-targeted strategies aims to provide important references for the research and development of cardiac-targeted drug delivery. |
