穿山龙提取物的降血尿酸作用及醇提工艺研究

摘要:
目的 研究穿山龙降血尿酸有效成分的提取工艺。方法 应用大鼠急性高尿酸血症模型评价穿山龙提取物的降血尿酸作用。以穿山龙中所含的薯蓣皂苷为指标,用正交试验法考察乙醇浓度、提取时间、提取次数和溶剂用量4种因素对提取结果的影响。薯蓣皂苷和血尿酸的含量均用HPLC测定。结果 穿山龙乙醇提取物具有降血尿酸作用。只有提取次数对穿山龙的薯蓣皂苷提取率有显著影响。结论 穿山龙降血尿酸有效成分的最佳提取工艺为:6倍量50%乙醇提取3次,每次1 h。

Abstract:
OBJECTIVE To optimize the extracting process of ethanol extracts with hypouricemic effect from Dioscorea Nipponicae Rhizoma. METHODS The hyperuricemia model of rats was utilized to evaluate the hypouricemic effect of ethanol extracts from Dioscorea Nipponicae Rhizoma. Orthogonal design was utilized to optimize the preparing conditions including the concentration of ethanol, amount of ethanol, extraction times and extraction duration per time when dioscin was selected as the evaluation index. Both dioscin and serum uric acid levels were measured by HPLC. RESULTS The ethanol extracts of Dioscorea Nipponicae Rhizoma had hypouricemic action. Only extraction times had significant effect on the content of dioscin. CONCLUSION The optimum extraction process was to extract three times using six-fold 50% ethanol and 1 h per time.
