摘要: |
目的 建立评价荜澄茄质量的GC指纹图谱分析方法。方法 采用水蒸气蒸馏法提取挥发油,GC测定14批荜澄茄药材。气相色谱条件:DB-Wax石英毛细管色谱柱(30 m×0.25 mm,0.25 μm),氢火焰离子化检测器(FID),进样口温度250 ℃,检测器温度250 ℃,柱温起始温度为50 ℃,以3 ℃·min-1升温至75 ℃,以10 ℃·min-1升温至130 ℃,以5 ℃·min-1升温至240 ℃,维持15 min。载气为氮气,流速为1.0 mL·min-1,进样方式为分流进样,分流比10∶1,进样量为1 μL。结果 依据14批药材的指纹图谱数据进行聚类分析,将样品分为2类,选定其中的12批优质样品建立共有模式,相似度分析结果表明,第Ⅰ类为优质药材,第Ⅱ类为一般品。结论 本法专属性强、重复性好,为科学评价与鉴定荜澄茄药材质量提供了依据。 |

Abstract: |
OBJECTIVE To establish the method of quality assessment and identification of GC fingerprints for Litsae Frctus. METHODS The essential oil from 14 batches of Litsae Frctus was extracted by steam distillation, and determined by GC. The measurement conditions applied were as follows: DB-Wax capillary column (30 m×0.25 mm, 0.25 μm), FID detector, inlet temperature was 250 ℃, the detector temperature was 250 ℃, and temperature of column was increased linearly from 50 ℃ to 75 ℃ at a speed of 3 ℃·min-1, from 75 ℃ to 130℃ at a speed of 10 ℃·min-1, from 130 ℃ to 240 ℃ at a speed of 5 ℃·min-1 (15 min). Carrier gas was nitrogen(1.0 mL·min-1). Split ratio was 10∶1. Volume was 1 μL. RESULTS The 14 samples were classified by 2 grades according to cluster analysis and the 12 superior in producing area samples were selected to establish the mutual models. The results of similarity analysis showed that the grade Ⅰ was commendatory and the grade Ⅱ was general. CONCLUSION This method is simple, quick and specific. It is reliable to evaluate the quality of Litsae Frctus. |
