摘要: |
目的 确定鳖甲多糖的最佳提取工艺,以苯酚硫酸法测定鳖甲多糖含量。方法 采用碱提法提取、醇沉得到鳖甲多糖;利用苯酚硫酸法显色,在490 nm处测定吸光度。结果 碱液浓度为8%、料液比为1∶25、提取温度为60 ℃、提取时间8 h为最佳提取工艺,最佳醇沉浓度为80%;平均回收率为100.2%,RSD为1.4%(n=9)。结论 本实验鳖甲多糖提取及含量测定操作简便、结果准确、重复性好,可用于鳖甲多糖的提取与含量测定。 |

Abstract: |
OBJECTIVE To study the optimum extration process of polysaccharides from Carapax Trionycis. A spectrophotometry method for content determination of polysaccharides from Carapax Trionycis was established with Phenolsulfuric acid coloration. METHODS Polysaccharides was extracted by NaOH solution and alcohol precipitation from Carapax Trionycis. The contents of po1ysaccharides from Carapax Trionycis were determined by spectrophotometry at 490 nm.RESULTS The results of orthogonal test in treatment of alkali extraction showed the optimum conditions were as the follows: NaOH concentration was 8%, treatment temperature was 60 ℃, extraction time was 8 hours, the ratio of the material and the solution was 1∶25. The average recovery rate was 100.23% and RSD was 1.237%(n=9).CONCLUSION The method is proved to be simple, accurate and reliable, and can be used to extract and determine the polysaccharides from Carapax Trionycis. |
