目的 建立鸟氨酸-α-酮戊二酸合剂中α-酮戊二酸含量测定的高效液相色谱法。方法 采用HPLC对鸟氨酸-α-酮戊二酸合剂中α-酮戊二酸进行定量测定,采用Hypersil BDS 色谱柱(4.6 mm×250 mm,5 μm),流动相为0.1 mol·L-1(NH4)H2PO4,pH为2.65,检测波长为215 nm,流速为1.0 mL·min-1。结果 α-酮戊二酸在0.28~280 µg内呈良好的线性关系(r =0.999 9),平均回收率为100.7%,RSD为1.5%(n=9)。结论 本法可准确、快速地定量检测鸟氨酸-α-酮戊二酸,可用于该制剂的质量控制。 |

Abstract: |
OBJECTIVE To establish an HPLC method for oxogluric acid of L-ornithine α-oxoglutarate. METHODS To establish a determination method of oxogluric acid in L-ornithine α-oxoglutarate by HPLC. Hypersil BDS column(4.6 mm×250 mm, 5 μm) was used and the mobile phase consisted of 0.1 mol·L-1(NH4)H2PO4 (pH 2.65). The detection wavelength was 215 nm and the flow rate was 1.0 mL·min-1. RESULTS Oxogluric acid showed a good linear relationship in the range of 0.28-280 µg (r=0.999 9). The average recovery of assay was 100.7% with corresponding RSD was 1.5%(n=9). CONCLUSION This mehod is accurate and fast, and can be used for the quality control of L-ornithine α-oxoglutarate. |
