转录因子FOXF1/F2/J1/M1/O1/O3/P3在肺腺癌中预后价值及免疫浸润相关性分析 |

摘要: |
目的 通过Forkhead box (FOX)转录因子家族在肺腺癌中的表达,阐明其对肺腺癌预后的影响。方法 通过GEPIA数据库分析FOX家族在转录水平的表达;通过Kaplan-Meier plotter数据库对FOX家族进行生存分析;通过cBioPortal数据库对FOX家族成员突变概率进行分析,并构建蛋白质互相作用网络;通过WebGestalt数据库对FOX家族成员进行通路富集分析;用Timer数据库对这些关键基因在肺腺癌中免疫浸润淋巴细胞分析。结果 GEPIA数据库显示FOXA1/A3/J1/M1/P3在肺腺癌的表达明显高于正常组织,而FOXF1/F2/O1/O3在肺腺癌中的表达明显低于正常组织(P<0.05)。FOXJ1/M1/P3 mRNA水平升高和FOXF1/F2/O1/O3 mRNA水平降低与肺腺癌总生存率显著相关(P<0.05)。肺腺癌的免疫浸润水平与FOXF1/F2/O1/P3表达相关。结论 FOXF1/F2/J1/M1/O1/O3/P3是肺腺癌预后不良的潜在标志物。FOXP3与肺腺癌的免疫浸润水平有关,可能参与肺腺癌中CD4细胞和B细胞的调节。 |

OBJECTIVE To elucidate the effect of Forkhead box(FOX) transcription factor family on the prognosis of lung adenocarcinoma through the expression of FOX transcription factor family in lung adenocarcinoma. METHODS The expression of FOX family at transcriptional level was analyzed by GEPIA database; the survival of FOX family was analyzed by Kaplan-Meier plotter database; the mutation probability of FOX family members was analyzed by cBioPortal database, and the protein interaction network was constructed; the pathway enrichment analysis of FOX family members was performed by WebGestalt database; and the immune infiltrating lymphocytes of these key genes in lung adenocarcinoma were analyzed by Timer database. RESULTS GEPIA database showed that the expression of FOXA1/A3/J1/M1/P3 was significantly higher than that of normal tissues, while the expression of FOXF1/F2/O1/O3 in lung adenocarcinoma was significantly lower than that of normal tissues(P<0.05). The increase of FOXJ1/M1/P3 mRNA level and the decrease of FOXF1/F2/O1/O3 mRNA level were significantly correlated with the overall survival rate(P<0.05). The level of immune infiltration in lung adenocarcinoma was correlated with the expression of FOXF1/F2/O1/P3. CONCLUSION FOXF1/F2/J1/M1/O1/O3/P3 is a potential marker for poor prognosis of lung adenocarcinoma. FOXP3 is related to the immune infiltration of lung adenocarcinoma and may be involved in the regulation of CD4 cells and B cells in lung adenocarcinoma. |
