摘要: |
目的 建立测定地黄寡糖中地黄苷A的高效液相色谱法。方法 采用Agilent TC C18色谱柱(4.6 mm×250 mm,5 μm),柱温35 ℃,流动相:甲醇-水(10∶90),流速1.0 mL·min-1;ELSD参数:漂移管温度110 ℃,载气流量3.2 L·min-1。结果 地黄苷A在0.2~1.2 mg·mL-1(r=0.999 3)内线性关系良好,平均回收率为98.8%(RSD=1.6%)。结论 该方法准确、简便、专属性好,可用于地黄寡糖中地黄苷A成分的含量测定,为地黄寡糖的质量控制提供了依据。 |

Abstract: |
OBJECTIVE To establish an HPLC method to determine the content of rehmannioside A in Rehmannia glutinosa oligosaccharide. METHODS The HPLC-ELSD was used with Agilent TC C18 column (4.6 mm×250 mm, 5 μm) at 35 ℃. The mobile phase was methanol and water(10∶90) with a flow rate 1.0 mL·min-1; The temperature of the drift tube in ELSD was 110 ℃ and the flow rate of dry air was 3.2 L·min-1. RESULTS The rehmannioside A had a good linear relationship between 0.2-1.2 mg·mL-1. CONCLUSION The method is accurate, simple with good specificity,and it could be used to determine the content of rehmannioside A in Rehmannia glutinosa oligosaccharide, which may be providing a evidence for the quality control of Rehmannia glutinosa oligosaccharide. |
