摘要: |
目的 探讨miR-9a对大鼠皮肤伤口愈合的影响及其作用途径。方法 构建大鼠伤口愈合模型,检测miR-9a在不同时间点皮肤伤口愈合组织中的表达水平。过表达miR-9a,利用原位杂交试验、HE染色、免疫荧光染色、划痕试验和伤口愈合试验分析差异。结果 与损伤前相比,损伤后7 d皮肤组织中miR-9a表达显著升高(P<0.001);过表达miR-9a组在损伤第14天和第21天损伤区域面积显著减小(P<0.05或P<0.001)。与对照组相比,转染miR-9a到角质形成细胞HaCaT后,EdU阳性细胞数量显著增加(P<0.001)、伤口愈合百分比显著升高(P<0.01)、迁移细胞的数量显著增加(P<0.001)。结论 miR-9a促进大鼠皮肤伤口愈合,其途径可能是通过促进角质形成细胞的增殖和迁移。 |

OBJECTIVE To investigate the effect of miR-9a on wound healing in rats, and to elucidate its mechanism. METHODS A rat wound healing model was constructed. The miR-9a expression in skin wound healing tissue was detected at different time points. Overexpression of miR-9a, differences between groups were analyzed by using in situ hybridization experiments, HE staining, immunofluorescence staining, scratch assays, and wound healing experiments. RESULTS Compared with pre-injury, miR-9a expression in skin tissue was significantly increased after 7 d of injury(P<0.001). In the miR-9a overexpressed group, the area of the injured area was significantly reduced(P<0.05 or P<0.001). Compared with the control group, after transfection of miR-9a into keratinocyte HaCat, the number of EdU-positive cells was significantly increased(P<0.001) at 14 d and 21 d after injury, the percentage of wound healing was significantly increased(P<0.01), and the number of migrated cells was significantly increased(P<0.001). CONCLUSION miR-9a promotes wound healing in rat skin by promoting proliferation and migration of keratinocytes. |
